A Punch Out!! sequel would be "fresh and new," says devs.

The devs for Punch Out!! Wii talked for a bit with Kotaku about their retro revived boxing title. Initially, Next Level Games considered having Princess Peach as a playable character. However the idea of hitting a woman senseless unless she falls was a hard pill to swallow on Nintendo's family friendly system.

Additionally, the thought of bringing Mario back as the referee was considered. But the toon shaded look of the game just wasn't a good fit for the 3D plumber. Instead, they opted for another Nintendo character to enter the ring, (Spoiler if you haven't played the game) the king of the jungle, Donkey Kong.

A bit of good news however is that the company talked about the possibility of a sequel. "We are not planning a sequel where simply the fighters are replaced," said Kensue Tanabe, one of the game's producers. Interesting choice of words! "But of course, Doc Louis must also be there!"
